background All of our liquid media and reagents are produced in a controlled environment designed to carefully control air pressure, temperature and particulate matter. The manufacturing area is a Class 10,000 environment with Class 100 laminar flow mixing, processing and filling areas. The manufacturing environment is monitored daily for particulate and microbial levels to ensure that the air handling system and cleaning protocols maintain a high standard of control. The water used in all production and testing is purified tissue culture water which is 18 megaohms/cm water purified by reverse osmosis, deionized through mixed bed resins and further filtered to 0.2 microns. All liquid media, sera and reagents are sterilized by filtration to 0.2 microns and sterility-tested according to the methods for sampling and testing described in the United States Pharmocopeia (Volume XXV, NF20). Each batch of liquid media is also tested for pH and osmolarity using routinely calibrated equipment.
The importance of pre-testing all reagents and components to ensure the success hematopoietic cell colony assays is self-evident. Our long standing experience in optimizing assay conditions has demonstrated the importance of careful selection and pre-testing of tissue culture supplies. As well as monitoring criteria such as colony size and cell viability, dishes have been tested for their inability to allow fibroblast attachment and growth which obscures or even inhibits hematopoietic colony formation. Double chamber culture slides have been tested for their suitability for collagen-based assays and use with cytochemical staining systems.
